Reduced Histone Expression or a Defect in Chromatin Assembly Induces Respiration.

چکیده
Regulation of mitochondrial biogenesis and respiration is a complex process that involves several signaling pathways and transcription factors as well as communication between the nuclear and mitochondrial genomes. Here we show that decreased expression of histones or a defect in nucleosome assembly in the yeast Saccharomyces cerevisiae results in increased mitochondrial DNA (mtDNA) copy numbers, oxygen consumption, ATP synthesis, and expression of genes encoding enzymes of the tricarboxylic acid (TCA) cycle and oxidative phosphorylation (OXPHOS). The metabolic shift from fermentation to respiration induced by altered chromatin structure is associated with the induction of the retrograde (RTG) pathway and requires the activity of the Hap2/3/4/5p complex as well as the transport and metabolism of pyruvate in mitochondria. Together, our data indicate that altered chromatin structure relieves glucose repression of mitochondrial respiration by inducing transcription of the TCA cycle and OXPHOS genes carried by both nuclear and mitochondrial DNA.

P-202: Reduced Expression of JMJD1A Histone Demethylase Gene in Testis Tissues of Infertile Men Referred to Royan Institute
Background: Epigenetic modifications are involved in different cellular processes through regulating chromatin dynamics. histone methylation is an important modification that can be dynamically regulated by histone methyltransferase and histone demethylase enzymes. JMJD1A (also known as JHDM2A and KDM3A) is a histone demethylase specific for H3K9me2/me1. JMJD1A is a key epigenetic regulator tha...
متن کاملP-217: Expression Analysis of The Histone Variant H2A.Z in Endometrium Tissue during The Menstrual Cycle
Background: The human endometrium undergoes cyclical changes including proliferation, differentiation, tissue breakdown and shedding (menstruation) throughout a woman’s reproductive life. The postovulatory rise in ovarian progestrone induces profound remodeling in chromatin structure of cells, and consequently differentiation of estradiol-primed endometrium. This change is crucial for embryo im...
متن کاملP-209: Decreased Expression of Histone Acetyltransferase CDY1 Gene in Testis Tissue May Lead to Decreased Expression of Transition Protein (TNP) and Protamine (PRM) Genes,Causing Male Infertility
Background: Infertility is a complex medical problem. About 15% of couples are infertile, and male infertility being involved in roughly 50% of the cases. Among these, many cases are associated with a severe impairment of spermatogenesis. During the last stage of spermatogenesis (spermiogenesis), sperm chromatin endures complex modifications in which histones are lost and depositioned with tran...
متن کاملINTRODUCTION Transcriptional activation of endogenous genes and of transgenes is influenced by the assembly of a repressive chromatin structure. Similarities between position effect variegation in Drosophila (Wakimoto, 1998) and variegated transgene expression in zebrafish
Transcriptional activation of endogenous genes and of transgenes is influenced by the assembly of a repressive chromatin structure. Similarities between position effect variegation in Drosophila (Wakimoto, 1998) and variegated transgene expression in zebrafish (Lin et al., 1994; Stuart et al., 1990) suggest that transgene expression is affected by chromatin assembly. To support this view, the h...
متن کاملKAT7/HBO1/MYST2 Regulates CENP-A Chromatin Assembly by Antagonizing Suv39h1-Mediated Centromere Inactivation
Centromere chromatin containing histone H3 variant CENP-A is required for accurate chromosome segregation as a foundation for kinetochore assembly. Human centromere chromatin assembles on a part of the long α-satellite (alphoid) DNA array, where it is flanked by pericentric heterochromatin. Heterochromatin spreads into adjacent chromatin and represses gene expression, and it can antagonize cent...
